KAT6A-CREBBP Fusion FISH probe

Gene Background: KAT6A codes for a zinc finger protein that regulates transcription via activation of RUNX1, while CREBBP encodes a ubiquitously expressed protein that monitors central cell cycle functions. The t(8;16)(p11;p13) rearrangement generates the KAT6A/CREBBP fusion, a rare but recurrent abnormality found in approximately 0.2-0.4% of acute myeloid leukemia (AML) patients. As of 2019, only about 100 cases of KAT6A/CREBBP-positive AML had been reported, mostly as single case reports. The abnormality is more common in therapy-related and neonatal AML.

Gene Summary
This gene encodes a member of the MOZ, YBFR2, SAS2, TIP60 family of histone acetyltransferases. The protein is composed of a nuclear localization domain, a double C2H2 zinc finger domain that binds to acetylated histone tails, a histone acetyl-transferase domain, a glutamate/aspartate-rich region, and a serine- and methionine-rich transactivation domain. It is part of a complex that acetylates lysine-9 residues in histone 3, and in addition, it acts as a co-activator for several transcription factors. Allelic variants of this gene are associated with an autosomal dominant form of cognitive disability. Chromosomal translocations of this gene are associated with acute myeloid leukemia.
